Tocopheryl Acetate

Tocopheryl acetate is a stabilized ester form of vitamin E used in cosmetics as an antioxidant and skin-conditioning agent.

What it is

Tocopheryl acetate is the ester of acetic acid and alpha-tocopherol, and is the most commonly used form of vitamin E in cosmetics. Esterifying the tocopherol blocks its reactive phenolic hydroxyl group, which makes the molecule more chemically stable and less prone to degradation than free vitamin E, giving formulas a longer shelf life. It functions as an antioxidant and skin-conditioning agent; the acetate group is thought to be cleaved after the ingredient is absorbed, releasing active tocopherol.

Safety & warnings

Tocopheryl acetate is generally well tolerated in topical products and is a long-established cosmetic ingredient with a favorable safety assessment from the European Commission's scientific committee. A trade-off is that, compared with free vitamin E, the acetate is more poorly absorbed and may deliver less of vitamin E's direct activity until it is converted in the skin. People with known vitamin E sensitivity should patch test, but irritation is uncommon.

Origin & sourcing

It is made by esterifying alpha-tocopherol with acetic acid. The tocopherol itself comes in two forms distinguished by their stereochemistry: a natural-source D-alpha (RRR) version and a fully synthetic DL-alpha version, with the prefix indicating origin. Vegetable oils are a common source of the tocopherol starting material.

History

Vitamin E was discovered in 1922 by Herbert McLean Evans and Katharine Scott Bishop, isolated in pure form in 1935, and first synthesized in 1938; the name tocopherol derives from Greek roots meaning to carry and to bear, with the -ol denoting an alcohol. The acetate ester was later developed as a more stable, longer-lived form of the vitamin for commercial use, blocking the phenolic hydroxyl group to reduce acidity and oxidation.
